Labs face a growing compliance burden and increased uncertainty due to new regulations surrounding laboratory-developed tests (LDTs). The U.S. FDA's final rule and Supreme Court's recent decision to overturn the Chevron doctrine are creating a complex regulatory environment that challenges the ability of public health labs to rapidly respond to emerging pathogens. This article explores the implications of these changes and more.

Chances are that these PCR plastics are viewed just as labware by most people. In this article we bring out how simple factors, such as PCR plate rigidity, color of the wells, and adhesiveness of plate seals, can affect your gene expression results and show why it is important to use quality PCR plastics.

Tumor metastasis is a complex process. It requires the ability of the cancer cells to invade their surroundings and travel to distant sites and survive. Dr. Traci Lyons studies the mechanism of COX-2 functioning in the metastasis of breast cancer cells. Find out how Dr. Lyons used Bio-Rad’s PrimePCR™ Assays to confirm COX-2 knockdown at the mRNA level.

Traci Lyons describes how PrimePCR Assays ended her laboratory’s struggle to find a qPCR assay that could reliably detect transcription of the cyclooxygenase-2 (Cox-2) gene, which they had identified as potentially prometastatic. Using the PrimePCR Assay for Cox-2, researchers in her lab were finally able to verify cyclooxygenase-2 knockdown, which is a necessary step toward confirming its role in breast cancer metastasis.

The significant drop in leprosy worldwide is attributed to the development of antimicrobial multidrug therapy (MDT). Resistance to rifampicin — the backbone of MDT treatment — has appeared. Controlling leprosy transmission requires routine surveillance for mutations in the drug target genes before, during, and after the course of treatment. New high resolution melt (HRM) assays offer leprosy researchers a faster, more economical method to investigate resistance targets.

Classifying and understanding genetic variation between populations and individuals is an important aim in the field of genomics. High resolution melt (HRM) analysis is the quantitative analysis of the melt curve of a DNA fragment following amplification by PCR and can be considered the next-generation application of amplicon melting analysis. Careful sample preparation and planning of experimental and assay design are crucial for robust and reproducible results.

Not all cDNA synthesis kits have the same capacity to protect the integrity of RNA. An optimal level of RNase inhibitor is critical for powerful RNase inhibition and efficient reverse transcription. The iScript™ cDNA synthesis kit comes with a 5x iScript reaction mix and iScript reverse transcriptase, which is also blended with a potent RNase inhibitor.
